As the highest difficulty Pandemonium Ruptures, completing Colossal Ruptures guarantees the spawn of a Realmwalker. Defeating it immediately unlocks Rift Portal leading to Deathtoll Chamber.
Deathtoll Chamber is a small, single-room dungeon. Clearing the monsters and completing the challenge at a high Torment level guarantees a Superior Lair Key drop.
Note that Blizzard has not explicitly defined which level qualifies as high, so IGGM.com recommends performing at least T10 or higher difficulty to ensure the highest drop rate; however, be sure to choose the highest difficulty that your build can complete quickly, otherwise it will have the opposite effect.

You only need to complete the event with a Mastery rating to have a chance to summon a Realmwalker: kill at least 200 enemies within the ritual circle before the time limit ends. Defeating Realmwalker whenever it appears will allow you to enter Deathtoll Chamber and obtain a key – the two paths lead to the same goal.
Belial Ambush Encounter
Encountering Belial Ambush during regular lair boss fights in Diablo 4 is a passive way to obtain extra Lair Keys.
When you use Greater Lair Keys to open the vault of a Greater lair boss, there’s a chance to trigger a Belial Ambush. Defeating them gives you a chance to obtain a Superior Lair Key. However, please note that since ambushes are entirely based on random probability, treat this strategy as an additional source of income, not a primary farming method.
New Method: Farming Glints of Hope Cache
Finally, a piece of new information: in the recent Diablo 4 3.1.1 Server Hotfix, the official announcement stated that Superior Lair Keys have now been added to the final repeatable Glints of Hope cache drop list.
